چکیده

A NEW FIELD in enzymology has emerged in the past decade with the discovery that RNA can act as an enzyme. First discovered in the cellular RNA-splicing and processing machinery in the form of self-splicing group 1 introns’ and precursor tRNA-processing RNase P (Ref. 2) RNA catalytic activity in a number of smaller RNAs has subsequently been identified3-5. The small, naturally occurring catalytic RNAs are generally found in the genomes of RNA viruses and in virus-related RNAs, which are believed to replicate by a rollingcircle mechanism. Recently, the crystal structure of one of these small catalytic RNAs, the hammerhead ribozyme, was elucidated by two research groups using different approaches. The first structure was that of the hammerhead ribozyme, in which the catalytic or ‘enzyme’ strand was composed of RNA and the RNA substrate was replaced with a ‘substrateanalogue’ strand composed of DNA. Ribozymes: structure and mechanism in RNA catalysis.

The hammerhead RNA is a small catalytic RNA found in a number of RNA virus genomes and virus-like RNAs. The recently determined crystal structures of hammerhead ribozymes reveal how a small RNA motif can fold up into a conformation suitable for mediating RNA cleavage.

Mechanisms of RNA catalysis.

Ribozymes are RNA molecules that act as chemical catalysts. In contemporary cells, most known ribozymes carry out phosphoryl transfer reactions. The nucleolytic ribozymes comprise a class of five structurally-distinct species that bring about site-specific cleavage by nucleophilic attack of the 2'-O on the adjacent 3'-P to form a cyclic 2',3'-phosphate. In general, they will also catalyse the r...

Mechanistic considerations for general acid-base catalysis by RNA: revisiting the mechanism of the hairpin ribozyme.

Several small ribozymes carry out self-cleavage at a specific phosphodiester bond to yield 2',3'-cyclic phosphate and 5'-hydroxyl termini. Prior mechanistic and structural studies on the HDV ribozymes led to the proposal that the pK(a) of C75 is shifted toward neutrality, making it an effective general acid. Recent mechanistic studies on the hairpin ribozyme have led to models in which protonat...

Ribozyme mechanisms and folding Ribozymes , the f i rs t 20 years

In 1982 we reported the first catalytic RNA or ribozyme : the self-splicing intron of the Tetrahymena pre-rRNA. Additional examples of natural ribozymes were soon found, and research in the field focused on their enzymic mechanism and secondary and tertiary structure. Ribozymes identified through in vitro selection extended the repertoire of RNA catalysis. Two directions of current and future i...

Metal ions: supporting actors in the playbook of small ribozymes.

Since the 1980s, several small RNA motifs capable of chemical catalysis have been discovered. These small ribozymes, composed of between approximately 40 and 200 nucleotides, have been found to play vital roles in the replication of subviral and viral pathogens, as well as in gene regulation in prokaryotes, and have recently been discovered in noncoding eukaryotic RNAs. All of the known natural...
